Decoding Cancer’s Circular Genome: 3D Insights into Extrachromosomal DNA with ec3D

Dr. Biswanath Chowdhury from the New York Genome Center will introduce ec3D, a pioneering method leveraging Hi-C data to reveal the 3D structure of extrachromosomal DNA (ecDNA) in cancer, exposing how these circular elements drive oncogene activation, therapy resistance, and tumor evolution by rewiring domain architecture and enabling enhancer hijacking—insights that are unattainable with traditional genomics approaches. This new 3D mapping approach not only uncovers hidden cancer vulnerabilities but also identifies “druggable” structural pockets in ecDNA, charting new paths for next-generation, precision therapies targeting the spatial organization of cancer DNA.
